摘要
目的观察撞伤Wistar大鼠肺微血管内皮细胞间连接成分血管内皮-钙粘附蛋白(VE-cadherin)的表达,了解撞击伤后肺微血管通透性的改变。方法利用荧光素钠法检测撞击伤后声门紧闭和开放状态Wistar大鼠肺组织的微血管通透性,动态观察撞击伤对细胞间连接的特有成分VE-cadherin表达的影响。结果撞击侧(右肺)肺微血管通透性均较伤前显著增高,各组在伤后同一时相点的值与正常对照组相比差异有统计学意义(P<0.01),声门紧闭组荧光素钠的含量高于声门开放组,差异有统计学意义(P<0.01)。胸部撞击伤后大鼠肺微血管内皮细胞VE-cadherin的表达降低,与对照组相比差异有统计学意义(P<0.01)。结论声门紧闭状态下撞击伤后肺微血管通透性高于声门开放状态,VE-cadherin在撞击伤后肺微血管内皮细胞中的表达下调,使得内皮细胞之间的连接疏松,通透性增加。
Objective To determine the expression of endothelial VE-cadherin in pulmonary microvascular endothelial cells in Wistar rats after lung impact injury for the alteration of pulmonary microvascular permeability. Methods The alteration of the microvascular permeability after blunt chest trauma (BCT) was examined by fluorescein sodium (FINa) method and by detecting the dynamic expression of endothelial VE- cadhefin in pulmonary microvascular endothelial cells. Results The content of FINa was significantly higher in the fight lung (the impact) than before injury (P 〈0.01), was higher at every time point after impact when compared with the normal control (P 〈0.01) , and also higher in the group of glottis closure than in the open one ( P 〈 0.01 ). The expression of VE-cadhefin in pulmonary microvascular endothelial cells was decreased after chest impact injury, with significant difference compared with the control group (P 〈0.01). Conclusion Closed glottis results in high microvascular permeability in the lung tissue than opened glottis in rats after blunt chest trauma. The decreased expression of VE-cadhefin in pulmonary microvascular endothelial cells further leads the loosed conjunctions between endothelial cells and subsequent increase of permeability.
