摘要
目的探讨候选抑癌基因OPCML(opioid-binding protein/cell adhesion molecule-like)在人乳腺癌组织中的蛋白表达及临床意义。方法应用免疫组化方法检测117例人乳腺癌组织、41例癌旁组织及24例乳腺腺病组织中的候选抑癌基因OPCML蛋白表达情况,结合临床病理资料进行统计学分析。结果 117例人乳腺癌组织、41例癌旁组织及24例乳腺腺病组织中OPCML蛋白的阳性表达率分别为19.66%(23/117)、90.24%(37/41)、70.83%(17/24),3种乳腺组织中的表达差异有统计学意义(P<0.05);在117例人乳腺癌组织中OPCML蛋白表达与组织学分级、淋巴结是否转移、pTNM分期显著相关(P<0.05);OPCML蛋白表达与Ki-67(r=-0.261)、HER-2(r=-0.294)、EGFR(r=-0.242)呈负相关关系(P<0.05)。结论候选抑癌基因OPCML在人乳腺癌组织中呈现低表达或表达缺失,且与Ki-67、HER-2及EGFR的表达呈负相关,可能在乳腺癌的发展中起抑癌作用。
[ Abstract ] suppressor gene, Objective To investigate the expression and clinical significances of the candidate tumor opioid binding protein/cell adhesion molecule-like (OPCML) gene in human breast cancer tissues. Methods Immunohistochemical assay was used to detect the expression of OPCML in 117 tissue samples of breast cancer, 41 samples of adjacent tissue and 24 samples of normal breast tissue. The resuhs were analyzed with clinical data. Results The positive rate of OPCML was 19.66% (23/117), 90.24% ( 37/41 ) and 70.83% (17/24) respectively in the breast cancer, adjacent tissue and normal breast tissue, with that in the breast cancer significantly lower than those in the other 2 tissues (P 〈 0.05 ). Significant correlations were found in the expression of OPCML with histological grade, pTNM staging and lymph node metastasis of breast carcinoma (P 〈 O. 05 ). Significantly negative correlations were seen among the expression of OPCML with that of Ki-67 (r= -0.261, P〈0.05), HER-2 (r= -0.294,P〈0.05) and GFR (r= -0.242, P〈0.05) respectively. Conclusion The candidate tumor suppressor gene OPCML is mildly expressed or deleted in the breast cancer, and its expression is negatively correlated with the expression of Ki-67, HER-2 and EGFR. It might play a role as suppressor gene in the development of breast cancer.
