摘要 目的检测抑癌基因PTEN与FHIT蛋白在乳腺癌和癌旁组织中的表达,分析其与临床病理特征的联系。方法应用免疫组化法检测68例乳腺癌,20例癌旁组织中PTEN和FHIT蛋白表达,分析其与乳腺癌临床病理特征的关系。结果在乳癌组织中,PTEN(χ2=8.351,P<0.01)与FHIT(χ2=9.748,P<0.01)均低表达,差异有统计学意义;淋巴结转移组标本,PTEN(χ2=6.338,P<0.05)与FHIT(χ2=7.215,P<0.05)均低表达,差异有统计学意义;分期较晚的标本中,PTEN(χ2=13.388,P<0.05)与FHIT(χ2=11.045,P<0.05)均低表达,差异有统计学意义。但二者在乳腺癌中表达无相关性。结论 PTEN和FHIT在乳腺癌中有较低的表达,显著低于癌旁乳腺组织。它们的低表达与乳腺癌的发生发展有一定生物学意义。它们在乳癌中表达无相关,提示这两种蛋白表达可能是相对独立的事件。二者可作为判断乳腺癌生物学行为和临床预后的参考指标。 Objective To investigate the expression and significance of PTEN and FHIT in breast cancer tissue and normal adjacent tissue, and the correlation with clinicopathological factors. Methods Immunohistochemistry (Elivision method) was used to detect the expressions of PTEN and FHIT in 68 issues of breast cancer and 20 of normal adjacent tissue. Results In the cancer tissues, the decreased expression of both PTEN ( X2 = 8.351, P 〈 0.01 ) and FHIT ( X2 = 9. 748, P 〈 0.01 ) can be seen, whose differences were of statistical significance ; in lymph node metastasis specimen, the decreased expression of both PTEN (X2 = 6. 338 ,P 〈 0.05 ) and FHIT( X2 = 7. 215 ,P 〈 0.05 ) can be seen, whose differences were of statistical significance;in the specimens from the later stage cancer, the decreased expression of both FFEN(X2 = 13. 388 ,P 〈 0.05 ) and FHIT(X2 = 11. 045, P 〈 0.05 ) can be seen, which the differences were of statistical significance. There was no correlation between the expressions of PTEN and FHIT in breast cancers. Conclusion The expression of PTEN and FHIT in breast cancers is lower than the normal adjacent tissues ,which has a possible biological significance in the occurrence and development of breast cancer. No correlation between the expressions of PTEN and FHIT in breast cancers suggests that the expressions of these two proteins may be relatively independent cases. The two proteins could be used as a reference index in determining the biological behavior of breast cancer and clinical prognosis.
