山楂叶悬钩子水提取物对转化生长因子-β刺激的肝星状细胞活化的抑制作用及其机制 被引量:1

Inhibiting effects and mechanism of aqueous extract of Rubus crataegifolius for activation of tHSC/Cl-6 stimulated by TGF-β

摘要 [目的]探讨山楂叶悬钩子水提取物(RC—H)对转化生长因子(TGF—β)刺激的肝星状细胞(HSC)活化的影响及其机制.[方法]采用MTT法测定RC-H对大鼠肝星状细胞(tHSC/C1—6)存活率的影响;采用TGF-β促活化tHSC/C1—6细胞,蛋白印迹分析RC-H对TGF-β刺激HSC信号转导的影响.[结果]0~160mg/LRC-H对tHSC/Cl-6存活率无显著影响(P〉0.05),320,640,1280mg/LRC-H显著抑制tHSC/C1—6的存活率(P〈0.05).40,80,160mg/LRC—H可抑制TGF-β活化tHSC/Cl-6细胞中α-SMA蛋白表达,下调TIMP-1蛋白表达,上调MMP-13蛋白表达,降低p-Erk和p-JNK蛋白表达,对p-p38无显著影响.[结论]RC—H可通过介导MAPK信号通路中Erk和JNK磷酸化而抑制TGF-β刺激的肝星状细胞活化. OBJECTIVE To study the effects and its mechanism of aqueous extract of Rubus crataegifoli- us (RC-H) for activation of hepatic stellate cells tHSC/C1-6 stimulated by transforming growth factor-β(TGF-β). METHODS Effects of RC-H on cell viability of tHSC/C1-6 were detected by MTT, and on the signal transduction of tHSC/C1-6 activated by TGF-β were detected by Western blot. RESULTS 0 160 mg/L of RC-H had no obvious effects for the cell viability of tHSC/C1-6 (P〉0.05), and 320, 640 and 1 280 mg/L of RC-H had significant effects for the cell viability of tHSC/C1-6 (P〈0.05). 40, 80 and 160 mg/L of RC-H in tHSC/C1-6 activated by TGF-β inhibited the expression of α-SMA and TIMP-1, increased the expression of MMP-13 and decreased the expression of p-Erk and p-JNK, and had no effect on p-p38. CONCLUSION Aqueous extract of RC-H might inhibit the activation of tHSC/C1-6 stimulated by TGF-β via phosphorylation of JNK and Erk in MAPK signal pathway.
