摘要
目的:建立测定香椿叶中山柰酚-3-O-鼠李糖苷含量的方法,为其质量控制提供参考。方法:采用高效液相色谱法。色谱柱为Thermo Hypersil ODS(150mm×4.6mm,5um),流动相为甲醇-水(55:45,V/V),流速为1.0ml/min,检测波长为254nm,柱温为35℃。结果:山柰酚-3-O-鼠李糖苷的质量浓度在5.14-51.40μg/ml范围内与其峰面积积分值呈良好的线性关系(r=0.9997);精密度、重复性、稳定性试验的RSD〈2%;平均加样回收率为96.77%~103.02%,RSD为3.75%~4.64%(n=3)。结论:该方法准确、重复性和稳定性好,可用于香椿叶中山柰酚.3.D.鼠李糖苷的含量测定。
OBJECTIVE: To establish a method for the content determination of kaempferol-3-O-rhamnoside in Toona sinensis, and to provide reference for quality control of it. METHODS: HPLC method was adopted. The determination was performed on Thermo Hypersil ODS (150 mm×4.6 mm, 5 μm) column with mobile phase composed of methanol-water (55:45, V/V) at the flow rate of 1.0 ml/min. The detection wavelength was set at 254 nm and column temperature was 35 %. RESULTS:The linear range of kaempferol-3-O-rhamnoside were 5.14-51.40μg/ml(r=0.999 7). RSDs of precision, reproducibility and stability tests were all lower than 2%, and average recoveries were from 96.77% to 103.02% (n=3). RSDs were 3.75%-4.64% (n=3). CONCLUSIONS: The method is accurate, reproducible and stable. It can be used for the content determination of kaempferol-3-O-rhamnoside in T. sinensis.
