摘要
目的 :研究胰岛素样生长因子 I(IGF- I)及其受体 (IGF— IR)基因在脐血淋巴细胞的表达以及与免疫调节、免疫发育的关系。方法 :通过 RT— PCR技术检测 IGFs基因的转录水平 ;利用反义寡核苷酸 (ASON)阻抑淋巴细胞IGF— IR m RNA的表达后 ,通过淋巴细胞增殖试验观察细胞免疫状况。结果 :脐血淋巴细胞有 IGF— I和 IGF— IRm RNA表达 ,经 IGF— IR ASON作用后不同程度地降低了淋巴细胞 IGF— IR m RNA的转录水平 ,同时淋巴细胞的增殖也被显著抑制。结论 :IGFs能够调节脐血 T淋巴细胞对 PHA的免疫应答 ,并可能参与新生儿的免疫发育 ,同时在维持新生儿细胞免疫功能方面具有重要的作用。
Objective:To study both the gene expression of insulin-like growth factor I(IGF-I),insulin-like growth factor I receptor(IGF-IR) in cord blood lymphocyte and the relation among IGFs,immune regulation and immune development.Method:We observed not only the transcription level of IGFs gene using reverse transcriptase polymerase chain reaction,but also cell immune condition by 3H thymidine uptake experiment after antisense oligonucleotide (ASON) inhibiting the expression of IGF-IR mRNA of cord blood lymphocyte. Result:There were the expression of IGF-I mRNA in cord blood lymphocytes.The level of IGF-IR mRNA decreased following exposure to ASON.The lymphocyte proliferation response was markly inhibited.Conclusions:IGFs can regulation immune response of T lymphocyte to PHA and may be involved in neonatal immune development and cell immune function.
