B7-H3在急性胰腺炎大鼠血清中的表达及意义 被引量:3

摘要 目的观察急性胰腺炎(acute pancreatitis,AP)大鼠血清协同刺激分子B7家族3(B7-H3)表达水平的变化及其与肿瘤坏死因子α(TNF-α)、胰腺病理学评分的关系,探讨其在AP发病过程中的作用及意义。方法健康雄性SD大鼠30只,随机分为5组:对照组,AP模型组(分为AP 6h组、AP 12h组、AP 24h组、AP 48h组),每组6只。酶联免疫吸附测定法测定血清B7-H3和TNF-α水平并分析两者表达水平变化的相关性;切取胰腺组织行胰腺病理学评分并分析其与B7-H3表达水平变化的相关性;全自动生化分析仪测定血清淀粉酶水平。结果 AP组血清中B7-H3、TNF-α和淀粉酶水平及胰腺病理学评分均显著高于对照组(P<0.01)。血清B7-H3与TNF-α水平和胰腺病理学评分变化均呈显著正相关(r=0.969、0.952,均P<0.01)。结论协同刺激分子B7-H3在AP血清中的表达水平与AP病情的严重程度有关,B7-H3可能通过影响TNF-α的产生而参与了AP的炎症反应过程。 Objective To observe the dynamic changes in the serum levels of the co-stimulatory molecule B7homology 3(B7-H3)in rats with acute pancreatitis(AP),and to analyze the correlation between B7-H3and tumor necrosis factor-α(TNF-α)and pancreatic histological scores,and to explore its role and significance in AP.Methods Thirty SD male rats were divided randomly into five groups:control group,acute pancreatitis group(AP 6h,AP 12h,AP 24h,AP 48h),each included 6rats.The serum B7-H3and tumor necrosis TNF-αconcentration were determined by enzyme-linked immunoabsorbent assay(ELISA),and the correlation between the expression level of them was analyzed.Pancreatic sections were stained with hematoxylin and eosin for histological analysis and scoring,and the correlation between the expression level of B7-H3and pancreatic histological scores was analyzed.Serum amylase was determined by automatic biochemical analyzer.Results The levels of B7-H3,TNF-αand amylase in serum and histological scoring of AP group were significantly higher than those of control group(P <0.01).The serum concentration of B7-H3and TNF-αand pancreatic histological scores were positively correlated(r=0.969,0.952,both P <0.01).Conclusion The serum concentration of B7-H3is correlated with the severity of AP,B7-H3probably plays a role in the inflammatory process of AP by the production of TNF-α.
