摘要
目的 研究子宫内膜癌患者外周血淋巴细胞染色体不稳定性 ,探讨子宫内膜癌患者外周血淋巴细胞染色体畸变 (CAR)、脆性部位 (fra)、姐妹染色单体交换 (SCE)、微核 (MN )、核仁形成区 (NOR)的相关性。方法 对 19例子宫内膜癌患者外周血培养 ,常规收获细胞 ,制片 ,分别对淋巴细胞染色体 CAR、fra、SCE、MN、NOR检测并进行相关分析。结果 119例子宫内膜癌患者染色体结构畸变以出现次数的多少排列顺序为 :3、2、1、5、7、17号染色体。 2染色体非二倍体、CAR、fra、SCE频率、MN率、Ag- NOR分别为 2 1.4%、(9.9± 3.5 ) %、7.8%、(7.2± 1.7)次 /细胞、(6 .8± 2 .3)‰、(6 .1± 1.4)个 /细胞。正常对照组为 15 .2、(4 .0± 1.9) %、3.1%、(5 .1± 0 .7)次 /细胞、(2 .2± 1.6 )‰、(5 .3± 1.0 )个细胞。 3染色体 CAR、fra、SCE、MN间相关性分析发现正常对照组CAR与 fra之间呈正相关 ,余各项之间无相关。而子宫内膜癌患者除了 CAR与 MN无相关外 ,各指标之间均呈正相关。结论 子宫内膜癌患者外周血淋巴细胞染色体存在着数目及结构畸变 ,染色体畸变是导致子宫内膜癌发生的重要原因。肿瘤患者淋巴细胞染色体不稳定性的发生是其必然的表现 ,通过对高危人群进行染色体不稳定性检测有助于筛查高危患者。
Objective In order to investigate chromosomal instability for lymphocytes of endometrial carcinoma (EC) and relationships of CAR,fra,SCE,MN,NOR in the peripheral blood lymphocytes in these patients.Methods Cytogenetic studies have been carried out using G-banding technique in peripheral blood lymphocytes of 19 patients of EC and 29 normal controls(NC).Results ①The chromosomal structural aberration of EC occurred on chromosome 3,2,1,5,7,17,in ± orders.②The frequency of chromosomal aneuploid in EC (21.4%) was significantly higher than in NC (15.2%) (P<0.05).The frequency of chromosomal aberration (9.9±3.5)%,SCE(7.2±1.7) times/cell and MN (6.8±2.3)‰ in the EC were significantly higher than (4.0±1.9)%,(5.1±0.7) times/cell,(2.2±1.6)‰ in the NC (P<0.001).The frequency of Ag-NOR in EC [(6.1±1.4)/cell] was not significant compared with NC [(5.3±1.0)/cell] (P>0.05).③The relationship of CAR,SCE,fra,MN in the NC,CAR versus fra had a positive correlation,the others had no relationships;while in the EC,CAR versus fra,CAR versus SCE,fra versus SCE,fra versus MN and SCE versus MN all had positive correlations,except CAR versus MN.Conclusion Our results suggest that patients with endometrial carcinoma show a certain degree of chromosomal instability that might be related to the predisposition to neoplasia,the presence of chromosomal instability further provides an important aid to clinical diagnosis of EC.
