系统性红斑狼疮患者外周血中Fas和sFas基因的表达

Expression of Fas and sFas Genes in the Peripheral Blood of the Patients with Systemic Lupus Erythematosus

目的 研究系统性红斑狼疮 (SLE)患者外周血中Fas和sFas基因的表达情况 ,探索细胞凋亡在SLE发病中的作用。方法 采用荧光标记的RT -PCR半定量法 ,以 β -actin为内参照 ,对 38例SLE患者和 2 5名正常健康人外周血白细胞凋亡相关基因Fas和sFasmRNA的表达水平进行检测。结果 SLE患者Fas和sFas基因的表达水平分别为 (4 .1± 1.2 )和 (1.6± 0 .5 ) ,与正常对照组 (2 .7± 0 .7)和 (0 .6 9± 0 .32 )相比有高度显著性差异 (P <0 .0 0 1)。结论 SLE患者外周血细胞凋亡相关基因Fas和sFas的表达异常 。

Objective To study on the mRNA expression of Fas and sFas genes in the peripheral blood of the patients with systemic lupus erythematosus(SLE). Methods Thirty-eight patients with SLE and 25 normal contral were selected and the mRNA expression of Fas and sFas genes related to the apoptosis of the peripheral white cells was analyzed by fluorescence-lebelled RT-PCR semi-quantitative method. Results The mRNA expression of Fas and sFas genes in 38 patients with SLE were (4.1±1.2) and (1.6±0.5) respectively and was significantly higher than that in 25 normal control (2.7±0.7) and (0.69±0.32), P <0.001.Conclusion The mRNA expression of Fas and sFas genes related to apoptosis in the peripheral blood white cells of the patients with SLE are abnormal and the apoptosis may play an important role in the pathogenesis of SLE.
