地龙对哮喘小鼠气道重塑的作用 被引量:19

摘要 目的探讨地龙对哮喘小鼠气道重塑的干预作用。方法 Balb/c小鼠40只,随机分成4组:模型组、空白组、地龙组、孟鲁司特钠组。腹腔注射卵清蛋白(OVA)20μg致敏,2.5%OVA雾化吸入激发建立哮喘小鼠模型。肺泡灌洗液(BALF)计数各炎性细胞;HE染色观察病理变化;Masson染色和PAS染色分别检测胶原沉积情况和PAS阳性细胞数,用Image-ProPlus 6.0系统定量。结果与空白组相比,模型组中炎性细胞数显著增加,胶原蛋白沉积和粘蛋白糖原表达显著提高;地龙组和孟鲁司特钠组BALF中嗜酸性粒细胞数(EOS)、气道胶原蛋白沉积、粘蛋白糖原表达明显低于模型组,差异有统计学意义。结论地龙可有效改善哮喘引起的气道重塑。 Objective To investigate the inhibitory effect of earthworm on airway remodeling in the murine model of chronic allergen-induced asthma. Methods Balb/c mice were randomized into four groups, including the asthmatic model group, the control group, the earthworm group and the Montelukast Soudium tablets group. To establish the chronic asthma model, mice were sensitized with 20 μg ovalbumin (OVA) by intraperitoueal injection and stimulated with atomization of 2.5% OVA. Mice were then sacrificed at 24 h after the last OVA inhalation and bronchoalveolar lavage fluid (BALF) was collected to count the inflammatory cells. Haematoxylin and Eosin (HE) staining was used for pathological examination. Masson Trichrome and Periodicaeid-Sehiff (PAS) staining were chosen for measuring collagen deposition and mucin-positive goblet cells in the lung tissue by using Image-ProPlus 6.0 syetem, respectively. Results Compared to the control group, the number of inflammatory cells, the airway collagen deposition and mucin glycogen expression were significantly increased in the model group. Compared to the model group, eosinophils, the collagen deposition and mucin glycogen expression were markedly decreased in both earthworm- and Montelukast Soudium tablets-treated groups. Conclusion Earthworm may improve the airway remodeling in asthma mice.
