摘要
目的 探讨焦炉工外周血淋巴细胞热应激蛋白 70 (HSP70 )水平与DNA损伤的关系。方法 应用Westernblot法测定淋巴细胞HSP70水平 ,用微核分析和慧星试验 (CometAssay)测定DNA损伤程度。结果 焦炉工组淋巴细胞HSP70水平 (196 0± 5 36 )高于对照组 (1712± 44 2 ) ,差异有显著性(P <0 .0 5 ) ;焦炉工组微核率 (5 16 2 /10 6± 2 2 0 5 /10 6)和DNA损伤积分 (6 70 0 0 /10 6± 6 4934/10 6)也都明显高于对照组 (2 36 2 /10 6± 12 41/10 6、2 8936 /10 6± 30 874/10 6) ,差异有显著性 (P <0 .0 5 )。焦炉工人组中淋巴细胞HSP70水平与微核率和DNA损伤积分均呈明显的负相关 ,且差异有显著性 (r值分别为- 0 .46 1、- 0 .6 6 3,均P <0 .0 1)。结论 焦炉工外周血淋巴细胞HSP70水平的增高可能与保护DNA免受致癌物损伤有关。
Objective To explore the relationship between HSP70 level and the degree of DNA damage in lymphocytes of workers exposed to coke oven emissions. Methods Level of HSP70 in peripheral blood lymphocytes was measured by Western blot,and the degree of DNA damage was detected by micronucleus and Comet Assay. Results Compared with the control group,HSP70 levels in the exposed group increased(1?960±536 vs . 1?712 ±442, P <0.05),and micronucleus rate(5?162/10 6±2?205/10 6 vs .2?362/10 6±1?241/10 6) and DNA damage score(67?000/10 6±64?934/10 6 vs .28?936/10 6±30?874/10 6) also increased significantly.However,in the exposed group,the levels of HSP70 had strong negative correlation with micronucleus rates( r =-0.461, P <0.01) and DNA damage scores( r = -0.663, P <0.01). Conclusion The increase of HSP70 level may be involved in protection of DNA from genotoxic damage induced by carcinogen.
