摘要
目的:研究穿龙薯蓣种子质量检验方法,为穿龙薯蓣种子检验规程和质量标准的制定提供依据。方法:参考国际植物种子检验规程和农作物种子检验规程的方法对不同产地穿龙薯蓣种子质量进行测定。穿龙薯蓣种子净度分析送检样本最少500 g,试验样本最少50 g;真实性检验采用种子外观形态比较;质量测定采用百粒重;含水量测定采用高温烘干法3 h;发芽前处理采用5℃低温层积20 d,然后用100 mg·L^(-1)的GA3溶液浸种30 h方法,种子发芽采用20℃恒温砂石培养床萌发;生活力采用TTC染色法,染色温度为45℃,TTC浓度为0.6%,染色时间为4 h。结果:穿龙薯蓣种子生活力和种子发芽率呈显著正相关,发芽率Y对生活力X回归方程为Y=-10.591+1.058X。结论:初步建立了穿龙薯蓣种子扦样,净度分析,真实性鉴定,质量,含水量,发芽及生活力7项指标的检验方法。
Objective: To study the quality inspection method of the seed of Dioscorea nipponica Makino, provide the basis for the development of testing procedures and quality standards. Methods: Refer to the international seed testing rules and the method of crop seed inspection procedures, test different regions seed quality content. Results: The submitted samples of D. nipponica seed is at least 500 g for purity analysis, and test samples at least 50 g; authenticity identification of seed by appearance shape comparison; quality determination by the hundred seed; water content determination by high temperature drying method for 3 h; before germination by 5 ℃ low temper-ature stratification about 20 d, and then use the method of 30 h 100 mg· L^-1 GA3 solution soaking, germination by 20 ℃ sand culture bed germination ; TTC staining method was used for seed viability test, the dyeing temperature was 45 ℃, TTC concentration was 0. 6% , dyeing time was 4 h. Seed germination rate(Y) was significantly correlated with seed viability( X), and the regression equation was Y = - 10. 591 + 1. 058X. Conclusion: The 7 indicators test meth-od of D. nipponica seed have been preliminary established. 7 indicators are : sampling, purity analysis, authenticity identification, quality, water content, germination rate and seed viability.
