摘要
[目的]探讨外源生长素IAA、赤霉素、水杨酸、茉莉酸甲酯和乙烯利对龙眼胚性愈伤组织(emberogenic callus,EC)生长素响应因子DlARF5-1基因表达的影响。[方法]以龙眼EC为试材,利用实时荧光定量PCR法检测龙眼EC生长素响应因子DlARF5-1在不同激素处理下的表达情况。[结果]结果表明,龙眼EC生长素响应因子DlARF5-1对不同浓度的生长素、赤霉素、水杨酸、茉莉酸甲酯和乙烯利都有响应。DlARF5-1基因在1.0 mg/L IAA处理下的表达量高于对照水平,经12.0 mg/L GA3、75μmol/L SA、75μmol/L MeJA和30 mg/L乙烯利处理后,DlARF5-1基因的相对表达量较高,但都低于对照的表达量。[结论]外源生长素IAA可上调龙眼EC生长素响应因子DlARF5-1基因的表达,而外源赤霉素、水杨酸、茉莉酸甲酯和乙烯利对DlARF5-1基因的表达起负调控作用。
[Objective] The aim was to discuss effects of auxin IAA, GA3, salicylic acid, methyl jasmonie acid and ethylene on lnngan auxin response factor DIARF5-1 gene expression.[Method] The experiment determined the DIARF5-1 gene expression in the treatment of different hormones by qRT-PCR (real-time reverse transcription PCR) method. [Result] The resuhs showed that longan auxin response factor DIARF5-1 responsed to different concentrations of auxin, gibberellin, salicylic acid, methyl jasmnnic acid and ethylene. Then DIARF5-1 gene had higher expression than/he control level in the longan embryn callus after the treatmen of 1.0 mg/L IAA. Ahhough the DIARF5-1 gene had higher expression under the/restments of 12.0 mg/L GA3,75 μmol/L SA,75 μmol/L MeJA and 30 mg/L ethylene, all tile relative expression were lower than the control. [Conclusion] Consequently, exogenous auxin IAA could raise tile expression of longan auxin response factor DIARF5-1 gene, but exogenous GA3, salicylic' acid, methyl jasmonic acid and ethylene negatively regulated the expressinn of DIARF5-1.
