摘要 酶定向进化是模拟自然进化过程、用于改造酶性质的一种新策略.在以易错PCR为代表的无性进化技术基础之上,有性进化技术的发展使酶定向进化更接近于自然进化过程.与此同时为解决巨大容量的突变文库与有限的筛选技术之间的矛盾,许多新的高通量筛选方法不断涌现,大大加快了酶定向进化技术的发展.文章从构建高质量的突变基因文库和选择有效的高通量筛选方法两方面,详细总结了近几年出现的新技术并对其应用情况进行介绍. Directed evolution of enzyme is widely used to change the characteristics of enzyme, which mainly mimics the progress of natural evolution. Based on the neuter evolution technology represented as error-prone PCR, the development of sexual evolution technology makes directed evolution of enzyme is closer to natural evolution. At the same time, in order to deal with the contradictions between mutant libraries and limited screening methods, a series of high-throughput screening methods appears, so that the development of enzyme directed evolution is promoted. This essay mainly summarized some recent new methods and applications about mutant gene libraries and high-throughput screening methods.
