摘要
目的探讨中国汉族人群载脂蛋白M(apolipoprotein M,ApoM)基因-778T/C单核苷酸多态性与2型糖尿病合并冠状动脉粥样硬化性心脏病(冠心病)的相关关系。方法采用聚合酶链反应-限制性片段长度多态性方法对238例糖尿病合并冠心病的患者及236例对照组进行检测,分析ApoM基因-778T/C多态的基因型和等位基因频率的分布情况。结果ApoM基因-788T/C多态三种基因型(TT型,TC型和CC型)在2型糖尿病合并冠心病组分布频率分别为73.9%、24.3%和1.7%,在对照组分布频率分别为86.0%、13.1%和0.9%;两组比较差异有统计学意义(P=0.001)。ApoM基因-788T/C多态T、c等位基因的频率在2型糖尿病合并冠心病组及对照组中分布分别为86.1%、13.9%和92.6%、7.4%,两组比较差异有统计学意义(P=0.001)。Logistic回归校正性别、年龄、体质量指数、吸烟、原发性高血压、高脂血症等2型糖尿病合并冠心病的易患因素后。ApoM基因-778T/C多态与2型糖尿病合并冠心病的发病仍存在相关关系(OR=2.0,95%CI:1.3~3.0)。结论ApoM基因-778T/C多态可能是中国汉族人群2型糖尿病合并冠心病患者发病的独立危险因子。
Objectives To explore the association between the apolipoprotein M (Apo M) gene -788T/C polymorphisms and type 2 diabetes mellitus with coronary heart disease in Chinese Han population. Methods A case-control study was conducted in 238 patients with type 2 diabetes mellitus complicated with coronary heart disease and 236 controls. Polymorphic genotypes were determined by polymerase chain reaction-restriction fragment length polymorphism. Results The genotype frequencies in Apo M gene -788T/C polymorphism conformed well with the Hardy-Weinberg equilibrium in both case and control groups. The genotype frequencies of TI', TC and CC of Apo M gene -788T/C polymorphism were 86.0%, 13.1% and 0.9% in the controls, 73.9%, 24.3% and 1.7% in patients with type 2 diabetes mellitus with coronary heart disease respectively. Allele frequency distributions ofT, C ofApo M gene -788T/C polymorphism were 86.1%, 13.9% and 92.6%, 7.4% in case and control groups respectively. There were significant differences in the genotype and allele distribution of Apo M gene -788T/C polymorphism between cases and controls (P=0.001). Logistic regression analysis with adjustments for other risk factors (i.e. gender, age, body mass index, smoking, hypertension, hyperlipidemia) revealed that the Apo M -788T/C allele carriers had correlation with the risk of type 2 diabetes mellitus with coronary heart disease (OR=2.0,95% CI: 1.3-3.0). Conclusions The Apo M -788T/C allele may be considered as a genetic risk factor for type 2 diabetes mellitus with coronary heart disease in Han population of China.
