摘要
目的:探讨新一代降血压药洛沙坦对血管的保护作用。方法:由高血压组(SHR)和正常血压对照组(WKY鼠)组成,其中SHR组再分为治疗组(予洛沙坦20mg/kg·d)和非治疗组,于八月龄测量鼠尾动脉压、外周血淋巴细胞内游离钙浓度及电子显微镜下血管指标。结果:与正常血压对照组比较,高血压鼠外周血淋巴细胞内游离钙浓度增高( P< 0.001),病理上表现有血管内皮细胞肥大(P<0.05),血管腔表面粗糙变形。而经洛沙坦治疗后与非治疗组比较,治疗组细胞内钙超负荷得到逆转(P<0.01),血压显著下降(P<0.001),血管内皮损伤得到修复(P<0.001),接近正常对照组水平。结论:洛沙坦能逆转高血压鼠的钙超负荷现象,在降低动脉压的同时,有效地保护血管内膜。
Objechve:To investigate the mechanisms of angitension Ⅱ receptor blockade in protection to endothelium of hypertensive rats. Methods : The investigation comprised normotensive controls (WKY) and spontaneous hypertensive group (SHR), which included losartan-treated(20mg/kg·d)and untreated group. Relevant index such as blood pressure, intralymphocytic free calcium and ultrastractural changes in mesenteric artery with transmission electronic microscope were obtained in eith-month old rats. Results: The volume density of endothe lial cells and concentration of calcium ([Ca2+ ]i)of SHR was significantly higher than that of WKY(P<0. 05 & P<0.001).With the treatment of losartan, SHR show certain decrease in [Ca2+]i(P<0.01), bloopd pres sure and volume density of endothelial cells(P<0.001). Conclusions: Losartan could effectly revert the calci um-overlad state of SHE. Accompanied with the antihypertensive effect, rehabilitation of endothelial injury was shown.
