围着床期血清差异蛋白质筛选 被引量:1

摘要 目的 初步筛选与着床相关的蛋白质.方法 分别采集因输卵管因素接受体外受精-胚胎移植(VF-ET)治疗的妇女胚胎移植后2 d和2周后患者血清,以2周后患者血清人体绒毛膜促性腺激素(β-HCG)为标准,β-HCG<100 U/L者为着床失败组,共28例.β-HCG>100 U/L者为着床成功组,共27例.利用2-DE电泳技术对比分离两组间蛋白质谱,分离两组胚胎移植后2 d血清中的差异蛋白质,以MALDI-TOF技术进行蛋白测序.结果 差异蛋白质经质谱测序结果为人结合珠蛋白/触珠蛋白前体,分子量为45861Da.结论 着床成功与失败患者围着床期血清蛋白质谱存在差异,差异蛋白测序证实为人结合珠蛋白/触珠蛋白前体;该蛋白质在早期的血清检测中有可能特异性地反映着床的成败;可进一步研究,做成相关抗体来检测患者血清中的含量,早期预测着床成功性. Objective To screen implantation related proteins. Methods Serum samples of patients with prob- lem of fallopian tubes and undergoing IVF-ET treatment were collected on the second day after getting ovum. Serum β-HCG level after 14 days of transplantation was assessed to define the experimental group ( unsuccessful implantation) and control group (unsuccessful implantation). 2-DE and MALDI-TOF test were used to examine differences between the successful and the unsuccessful group, Results One protein, haptoglobin precursor, was found to have differences. Conclu- sion Further study of the protein function is needed and may lead to identification of a new biomarker of implantation.
