摘要 目的BALB/C小鼠受60Coγ射线不同剂量照射后,探讨不同剂量组以及各剂量组照后不同时间点小鼠血液及肝组织中胰岛素生长因子1受体(IGF-IR)基因表达的水平。方法利用实时荧光定量技术,采用60Coγ射线全身照射BALB/C小鼠,照射剂量为1.0、2.0、4.0 Gy,于照后6小时、照后12小时、照后24小时分别提取小鼠血液及肝脏组织样品中的RNA,观察比较IGF-IR的表达变化情况。结果不同剂量照后不同时间点,小鼠血液及肝脏样品中IGF-1R和β-actin的溶解曲线均为单峰,均为特异性扩增产物;IGF-1R在小鼠血液及肝脏组织中的表达趋势总体为下调,其中血液中4.0 Gy照后24小时下调表达量最低,肝脏组织中4.0 Gy照后6小时下调表达量最低。 This study is to investigate the irradiation effect at different does and different time on the expres-sion of insulin like growth factor type 1 receptor type 1 (IGF-1R) in mice .The mice were irradiated with 60 Co gamma rays at 1 .0 ,2 .0 and 4 .0 Gy ,then extracted total RNA from blood cells and liver tissues after irradi-ation 6 ,12 and 24 hours .IGF-1R expression in blood cells and liver tissues were detected by real-time fluo-rescent quantitative PCR .Solution curve ofβ-actin gene and (IGF-1R) with different doses and time was the single peak ,which means the specific ampliations . IGF-1R gene expression in blood cells and liver tissues showed down regulation ,The 4 .0 Gy 24 hours group expressed the lowest level in blood cells .The 4 .0 Gy 6 hours group expressed the lowest level in liver tissues .
