摘要
生防菌株B29是一个能够防治多种植物病害并促进植物生长的专利菌株,经鉴定为枯草芽孢杆菌。根据抗菌物质基因大小,本研究将提取的枯草芽孢杆菌基因组DNA采用Sau3AI部分酶切,回收3~5 kb酶切产物,将产物与经BamHI单酶切并经去磷酸化后的pUC19质粒连接,转化DH5α大肠杆菌,培养后共获得11 090个阳性克隆,按99.99%的概率计算,构建的基因组文库共覆盖了B29基因组3次,符合文库构建要求。该研究为生防菌株防治作物真菌病害及筛选抗菌物质调控基因奠定了基础。
Biocontrol strain B29 is a patented strain that can control a variety of diseases of plants and promote the growth of plants. It was identified as Bacillus subtilis. The genome DNA of B29 was digested by restriction enzyme Sau3AI. The products of digestion between 3 and 5 kb were collected according to the size of antimicrobial substances gene. The products after collection were connected with the plasmid pUC19 that was dephosphorylated and digested by restriction enzyme BamHI. 11,090 clones were obtained after cloning through Escherichia coli DH5α. Therefore,the genome of B29 was covered for three times by the genomic library just constructed if the probability is 99. 99%. And then it conformed to the requirements of library construction. This study supports the fungal disease prevention of plants by biocontrol stains and screening of regulated genes of antimicrobial substances.
