摘要 目的研究羟基红花黄色素A(HSYA)对小鼠3T3-L1前脂肪细胞增殖和分化的影响。方法采用MTT法、甘油三酯测定、RT-PCR和双萤光素酶报告基因检测等方法观察HYSA对3T3-L1细胞增殖和分化的影响及其可能的机制。结果 0.01 mg/L^10 mg/L HSYA在4 h^96 h抑制3T3-L1细胞的增殖,120 h促进细胞增殖,并具有浓度依赖性。HSYA减少3T3-L1细胞分化过程中脂质和甘油三酯的含量,增加HSL mRNA的表达和启动子活性。结论HSYA对3T3-L1细胞增殖的影响具有双向性,并通过促进HSL启动子的活性,增加mRNA表达,抑制其分化。 Objective To study the effect of bydroxysafflor yellow A (HSYA) on the proliferation and differentiation of mouse 313 - L1 preadipocytes. Methods The effect of HSYA on the proliferation and differentiation of 3T3 - L1 cells and possible mechanism were studied by using MTT assay, triglyceride assay, RT - PCR and dual luciferase reporter gene methods. Results HSYA level between 0.01 mg/L N 10 mg/L can inhibit the proliferation of 333 - L1 cells in 4 h - 96 h. However, HSYA conversely promoted the proliferation of cells at the time point of 120 h in a dose dependent manner. HSYA can significantly reduce the amount of lipid and triglyceride content, while increase HSL mRNA expression and promoter activity in differentiated 3T3 - L1 adipocytes. Conclusion HSYA can regulate the proliferation of 3T3 - L1 cells with a two - way mode, inhibit the 3T3 - LI cells differentiation and reduce the lipid content by promoting HSL promoter activity and mRNA expression.
