摘要
目的研究miR-205对人皮肤黑素瘤细胞系A375的侵袭能力和MMP2,MMP9的影响,探讨miR-205影响黑色素细胞侵袭的机制。方法将miR-205 mimics用阳离子脂质体LipofectamineTM2000转染A375细胞,Transwell侵袭实验检测细胞侵袭能力变化,Western blot和ELISA检测MMP2和MMP9蛋白表达变化。结果 Transwell侵袭实验显示转染miR-205 mimics后细胞的穿膜数(75.21±11.32)明显低于空白对照组(133.20±8.69)及阴性对照组(128.90±7.03),Western blot和ELISA结果均证实miR-205 mimics能明显减低MMP2/9蛋白表达,以上差异均有统计学意义(P均<0.05)。结论 miR-205可能通过减低MMP2和MMP9表达抑制黑素瘤细胞的侵袭能力。
Objective To investigate the effects of miR-205 on invasion and MMP2 and MMP9 expression in human skin malignant melanoma cell lines A375 and further elucidate its action mechanisms. Methods miR-205 mimics were transfected A375 by lipofectamine package. Cell invasion was studied by transwell invasion as- say. MMP2 and MMP9 proteins expression were investigated by western blot and ELISA. Results Transwell invasion assay demonstrated the number of A375 cells in miR-205 mimics group was significantly lower (75.21 + 11.32) than that in control group (133.20 + 8.69) and negative control group (128.90 + 7.03 ) at 48h after transfeetion. Western blot and ELISA confirmed that miR-205 mimics could significantly inhibit the expressions of MMP2/9 protein and secreting. Conclusion miR-205 inhibit the invaion of human skin ma- lignant melanoma cell may be through reduce the expression of MMP2 and MMP9.
