摘要
目的检测β-catenin在不同年龄人群非曝光部位皮肤组织中mRNA和蛋白的表达水平,初步探讨β-catenin在人皮肤年代老化过程中的意义。方法收集62例急腹症手术患者非曝光部位的皮肤标本,根据年龄分为三组:即青年组(<20岁)、中年组(30~45岁)和老年组(>65岁),用RT-PCR及Western blot检测每例皮肤组织中β-catenin mRNA和蛋白的表达量,分析不同年龄组之间β-catenin表达水平的差异。结果青年组、中年组和老年组β-catenin/GAPDH mRNA的表达水平分别是0.603 8±0.012 1,0.513 9±0.015 4和0.256 2±0.011 4,三组间比较的差异均有统计学意义(P<0.01),青年组与中年组,青年组与老年组,中年组与老年组比较差异均有统计学意义(P均<0.01);青年组、中年组和老年组β-catenin/GAPDH蛋白的表达水平分别是0.631 5±0.013 5,0.548 6±0.012 9和0.327 4±0.021 9,三组间比较差异均有统计学意义(P<0.01),青年组与中年组,青年组与老年组,中年组与老年组比较差异均有统计学意义(P均<0.01)。结论随着年龄的增长β-catenin的表达水平随之减少,推测人皮肤年代老化可能与β-catenin的表达下调有一定的关联性。
Objective To investigate the expression of β-catenin in unexposed skin of different ages,and to preliminary discuss its significance during chronological aging of human skin. Methods Sixty-two non-exposure skin sample were collected from the acute abdomen patients with different ages. The samples were divided into three groups : youth group( 〈 20-year-old ), middle age group ( 30 - 45-year-old ) and old age group ( 〉 65- year-old). The expression of β-catenin mRNA and protein of every case was detected by RT-PCR and West- ern blot. Results The mRNA ratio of [3-catenin/GAPDH was 0. 603 8 + 0. 012 1 in youth group,0. 5β 9 _+ 0. 015 4 in middle age group and 0. 256 2 -+0.011 4 in old age group;There were significant difference in β- catenin mRNA ratio expression among the three groups (P 〈 0.01 ). Obvious differences were observed in youth group compared with middle age group,unior group compared with old age group and middle age group compared with old age group( P 〈 0.01 respectively). The protein ratio of β-catenin/GAPDH was 0. 631 5 + 0.0β 5 in youth group,0.548 6 -+0.012 9 in middle age group and 0. 327 4 -+0.021 9 in old age group. There were significant difference in β-catenin proetein ratio expression among the three groups( P 〈0.01 ). Obvious differences were observed in youth gt^oup compared with middle age group, youth group compared with old age group and middle age group compared with old age group( P 〈 0.01 respectively). Conclusion The mRNA and protein expressions of β-catenin is decreased with the increase of age. This suggested that the decrease of expressions of β-catenin might have some correlation with chronological aging of human skirl.
