摘要
目的观察骨宁丸对兔骨关节炎模型的软骨光镜和扫描电镜下组织形态学的影响。方法成年新西兰兔35只,随机分为正常组、模型组、中药组、硫酸氨基葡萄糖组4组,以膝关节腔注射木瓜蛋白酶法建立膝骨关节炎模型。用药8周后,处死动物,取材观察形态学改变。结果模型组光镜和扫描电镜下均出现软骨退行性改变。光镜下观察,中药组和硫酸氨基葡萄糖组均有软骨损伤,但较模型组为轻,Mankin组织学积分分别为8.6±1.1和8.2±0.8,两组差异无显著性。扫描电镜下观察,中药组和硫酸氨基葡萄糖组均出现软骨表面的裂隙、小孔甚至浅溃疡,但表现较模型组为轻。结论骨宁丸对兔骨关节炎模型软骨退行性改变有治疗作用。
Objective To observe the effect of Bonening pill on histomorphometry of cartilage in a rabbit model of osteoarthritis with an optical microscope or/and an electron microscope .Methods Thirty-five adult New Zeal-and rabbits,were randomly divided into control group ,model group,traditional Chinese medicine group and glucosamine sulfate group .Osteoarthritis models wre established by intra-articular injection of papain .After 8 weeks of treatment ,rab-bits were executed ,and the morphological changes were observed .Results There were cartilage degeneration appeared in model group both in optical microscope and in scanning electron microscope .The optical microscope showed cartilage injury both in traditional Chinese medicine group and in glucosamine sulfate group ,the same as to model group .Mankin histological score was 8.6 ±1.1 and 8.2 ±0.8 respectively.There was no significant diversity between the two groups . The scanning electron microscope showed cracks ,holes and even shallow ulcer in the cartilage surface in Traditional Chi-nese medicine group and glucosamine sulfate groups , but they performed better than the model group .Conclusion Bonening pill has a therapeutic effect on degeneration of cartilage in rabbit osteoarthritis .models.
