摘要
目的探讨生黄合剂对大鼠心肌缺血再灌注损伤(MIRI)的干预作用。方法将48只SD大鼠随机分为6组,即空白对照组、模型组、阳性药物组、生黄合剂低、中、高剂量组,每组8只,分别给药7 d后,建立MIRI模型。观察各组大鼠血清肌酸激酶(CPK)、天门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)及心肌组织肿瘤坏死因子-α(TNF-α)水平的变化;用TUNEL法检测心肌细胞凋亡情况。结果与空白对照组比较,模型组血清CPK、AST、LDH及心肌组织TNF-α活性和心肌细胞凋亡指数明显升高(P<0.05);与模型组比较,各用药组能明显降低大鼠血清CPK、AST、LDH及心肌组织TNF-α含量和心肌细胞凋亡指数(P<0.05),其中以生黄合剂高剂量组作用较为明显。结论生黄合剂对大鼠MIRI具有明显的保护作用。
Objective To investigate the protective effects of Sheng-Huang mixture on myocardial ischemia-reperfusion injury (MI- RI) in rats. Methods SD rats (n =48) were divided randomly into six groups: normal control, model, positive control, low-dose, medi um-dose and high-dose Sheng-Huang mixture groups ( n = 8). After the drug was administered to the above 6 groups respectively seven days on end, the MIRI in rats was established. The content of lactic acid dehydrogenase (LDH), aspartate aminotransferase (AST), Creatine Ki nase (CK) in serum and tumor necrosis factor (TNF)-α in myocardial were detected. TUNEL was used to detect the myocardial apoptosis index. Results Compared with normal control group, the content of LDH, AST, CK in serum and TNF-α in myocardial and the myocardial apoptosis index of model group were increased obviously(P 〈 0.05 ). Compared with model group, the content of LDH, AST, CK in serum and TNF-α in myocardial and the myocardial apoptosis index of positive control group, low-dose, medium-dose and high-dose Sheng-Huang mixtrue groups were decreased obviously (P 〈 0. 05), above indexes in high dose Sheng-Huang mixtrue group were decreased much lower. Conclusions Sheng-Huang mixture has the protective effects on MIRI in rats.
