The relationship of IgG expression with OPN, MMP-2, MMP-9 and P16 in human glioma

摘要 目的检测IgG在人脑胶质瘤中的表达及其与OPN、MMP-2、MMP-9、P16表达的关系。方法免疫组化检测IgG、OPN、MMP-2、MMP-9、P16在80例胶质瘤组织中的表达,原位杂交和RT—PCR检测IgG mRNA在组织和胶质瘤细胞系U373细胞中的表达。结果胶质瘤组织和U373细胞系细胞中有IgG蛋白及IgG mRNA表达,IgG、OPN、MMP-2、MMP-9和P16的表达阳性率在不同级别胶质瘤组均有显著性差异,IgG与OPN、MMP-2、MMP-9的表达呈正相关,与P16表达呈负相关。结论胶质瘤中IgG、OPN、MMP-2、MMP-9高表达,P16低表达,可能在胶质瘤的发生发展中起一定作用。 Objective To investigate the expression of IgG and its relationship with OPN, MMP-2, MMP-9 and P16 in htunan glioma. Methods The expressions of IgG, OPN, MMP-2, MMP-9 and PI6 were detected by immunohistochemistry. IgG mRNA was detected with in situ hybridization in tissue, and with RT-PCR in U373 cells. Results IgG protein and IgG mRNA were expressed positively in glioma and U373 cells. There were significant differences for the positive rates of IgG, OPN, MMP-2, MMP-9 in glioma tissues between different grades. There was a positive correlation between IgG expression and those of OPN, MMP-2 and MMP-9, with negative correlation between IgG expression and that of P16. Conclusion Overexpressed IgG, OPN, MMP-2 and MMP-9, and downexpressed PI6 in glioma indicating that they may be involved in pathogenesis of glioma.
