摘要
目的探讨在小鼠内耳发育过程中,钙结合蛋白(CB)在内耳耳蜗内的表达是否存在时限性。方法获取C57BL/6小鼠E8开始至出生后一直到成年的内耳标本:E8,E9,E10,E12,E14,E15-E21,D1-D21,成年,进行冷冻切片,进行CB的荧光免疫组织化学染色并观察其在内耳表达的变化情况。结果在C57BL/6小鼠,除了在E16可辨认出螺旋神经元有CB部分表达外,以后直到成年都没有CB的染色表达,而在Scarpa神经元则从E8开始就有表达,其从E19开始先在内毛细胞开始有表达,逐渐在内外毛细胞都有表达,直到成年时表达消失。结论 CB在C57BL/6小鼠内耳耳蜗的表达具有时限性,因而可推测出对CB行免疫组织化学染色观察能有效鉴别C57BL/6小鼠及其他动物的再生毛细胞和原有毛细胞,但目前尚无法解释CB一些表达特点的相应机制。
Objective To explore the timeliness of Calbindin expression during the inner ear development of C57BIM6J mice. Methods We obtained inner ear specimens from precise pregnancy C57BIM6J mice (E8, E9, E10, E12,E14, E15 -E21 ) and postnatal mice (D1 - D22). All of the specimens were eyro-sectioned and marked by the Calbindin antibody. The distribution of Calbindin was observed during the inner ear development under a fluorescence microscope. Results In the spiral ganglion neurons, the expression of Calbindin only existed at El6 and then disappeared, but the expression began at E8 and continued until adulthood in Searpa ganglion neurons. The expression of Calbindin began at inner hair cells from El9, step by step diffused to both inner hair cells and outer hair cells over time. However, it disappeared after birth. Conclusions The timeliness of Calbindin expression during inner ear development of C57BL/6J mice is confirmed~ Calbindin is suggested to be useful for distinguishing regenerative hair cells from original hair cells in C57BL/6J mice. However, we can not explain the mechanisms of some Calbindin expression characteristics during inner ear development of C57BIM6 mice.
