生长分化因子15在子痫前期胎盘滋养细胞中的表达及意义 被引量:2

摘要 目的探讨生长分化因子-15(GDF-15)在子痫前期患者胎盘滋养细胞中的表达,探讨其在子痫前期发病机制中的作用。方法选择2009年12月至2010年10月在青岛市市立医院住院分娩的子痫前期孕妇140例,其中早发型轻度子痫前期孕妇35例(早发轻度组),晚发型轻度子痫前期孕妇35例(晚发轻度组),早发型重度子痫前期孕妇35例(早发重度组),晚发型重度子痫前期孕妇35例(晚发重度组);另选同期健康孕妇35例为对照组。采用RT-PCR检测胎盘滋养细胞中GDF-15 mRNA的表达量;采用免疫组化方法检测胎盘滋养细胞中GDF-15蛋白的表达。结果 RT-PCR显示子痫前期各组胎盘滋养细胞中GDF-15mRNA表达水平均高于对照组,差异有统计学意义(P<0.05);但子痫前期各组间分别比较,差异无统计学意义(P>0.05)。免疫组化显示GDF-15表达于胎盘滋养细胞的细胞浆和细胞外基质中,并在各组胎盘滋养细胞中均表达。子痫前期各组胎盘滋养细胞中GDF-15蛋白表达水平均高于对照组组,差异有统计学意义(P<0.05);但子痫前期各组间分别比较,差异无统计学意义(P>0.05)。结论 GDF-15在子痫前期胎盘滋养细胞中表达升高,GDF-15的表达水平变化可能与子痫前期发病有关。 Objective: To examine the relevance of the growth differentiation factor 15 with pathogenesis of preeclampsia (PE). Methods : A total of 140 pregnant women with PE and 35 normal pregnant women who were hospitalized in Qingdao Municipal Hospital from Dec 2009 to Oct 2009 were selected as the subject. One hundred and forty pregnant women with PE were divided into four groups : early mild PE group ( n = 35 ), late mild PE group ( n = 35 ), early severe PE group ( n = 35 ), late severe PE group ( n = 35 ), and thirty normal pregnant women were chosen as normal group for comparing. Semi - quantitative Reverse transcription (RT) PCR and Immunohistochemistry were used to investigute the expression levels of GDF - 15 mRNA and protein in placenta. Results: The results of RT - PCR showed that, compared with the normal pregnancy group, the expressions of GDF - 15 mRNA in PE group were significantly higher ( P 〈 0. 05 ) ; however, there were no significant difference among PE groups ( P 〉 0. 05 ). Immunohistochemistry revealed that the GDF - 15 protein expressed on the cytoplasm and extracellular matrix of the trophoblasts in all the groups of pre- eclampsia and normal group. The expression level of GDF - 15 protein in placenta in all the groups of PE were markedly higher than that of normal pregnancy group ( P 〈 0. 05 ) , while there were no difference among PE groups ( P 〉 0. 05 ). Conclusions : The increased expression lever of GDF - 15 in PE may be the factor closely related to pathogenesis of PE.
