制革污泥中产脂肪酶真菌的筛选及产酶条件优化 被引量:5

摘要 通过利用罗丹明B培养基和溴甲酚紫培养基初筛和酶活的测定法复筛,从制革厂含铬污泥(2.77mg/L)中筛选出一株产脂肪酶的真菌,经形态学观察、生理生化测定和分子生物学鉴定,初步鉴定该株菌为子囊菌门煤炱目下Teratosphaeriaceae科的一种,暂定名为Teratosphaeriaceae Cr12.同时对该菌株的摇床培养产酶条件进行了初步优化,得到最佳发酵条件为初始pH值为5.5,培养温度为35℃,种龄18h,接种量为2.5%(V/V),发酵周期为72h,转速为150r/min,酶活力最高达到8.5U/mL. aeriaceae under Capnodi ales of Ascomycota, was screened from tanning sludge in south China by the color agar plate method, morphological observation, determination of enzyme activity, physiological and bi- ochemical determination and molecular biological identification, and was nominated as Ter- atosphaeriaceae Crl2. Preliminary optimization of the shaker-based fermentation conditions of the lipase from Teratosphaeriaceae Cr12 were determined.
