摘要 对葡萄汁有孢汉逊酵母(Hanseniaspora uvarum)β-葡萄糖苷酶(β-glucosidase,HuBG)的分离纯化方法进行了研究。H.uvarum酵母发酵液经过硫铵沉淀、苯基疏水、离子交换、分子筛纯化后,得到电泳纯的β-葡萄糖苷酶。研究结果表明,野生酵母的β-葡萄糖苷酶在细胞中,大部分被分泌到胞外;酶的纯化倍数为34.88倍,比酶活为5.58 U/mg,SDS-page检测此酶的分子量约为80 kDa。 The isolation and the purification of β-glucosidase from Hanseniaspora uvarum in grape juice were investigated. In the experiment, H. uvarum fermenting liquid was treated by ammonium sulfate precipitation, hydrophobic interaction, ion exchange and gelfiltration chromatography to obtain HuBG through electrophoretic homogeneity. The experimental results revealed that the extracellular fraction (supematant) contained the majority of β-glucosidase and HuBG was purified to 34.88-fold with a specific activity of 5.58 U/mg. Furthermore, the molecular weight of BG1 was estimated to be 80 kDa by SDS-PAGE.
