青年乳腺癌中同源异型盒基因A5表达的研究

摘要 目的:探讨青年乳腺癌组织中同源异型盒基因(HOX)A5mRNA及其蛋白表达,研究乳腺癌肿瘤组织临床病理参数与其关联性.方法:运用TaqMan实时荧光定量逆转录聚合酶链反应(Real-time RT-PCR)技术检测120例青年乳腺癌(其中96例浸润性导管癌,原位癌16例、腺癌4例、硬癌4例)和96例青年乳腺良性病变中HOXA5mRNA表达,分析HOXA5基因表达强度与乳腺癌肿瘤原发灶组织临床病理参数间的相关性.结果:HOXA5 mRNA在乳腺癌肿瘤组织中相对表达量0.82 ~ 189.06,平均为20.69±29.15,乳腺良性病变中相对表达量5.84~80.72,平均为31.05±17.06;乳腺癌HOXA5mRNA表达显著低于乳腺良性病变(P<0.01).乳腺癌伴腋窝淋巴结转移阳性患者HOXA5mRNA表达强度较淋巴结未转移者显著降低(P<0.01).HOXA5mRNA表达与乳腺癌肿瘤原发灶大小、临床分期、组织学分型未见相关性(均P>0.05).结论:青年乳腺癌肿瘤原发灶组织细胞中HOXA5mRNA表达强度较乳腺良性病变组织显著下降.HOXA5mRNA表达明显降低与乳腺癌腋窝淋巴结癌转移关联性较大. Objective:To explore the expression of homeobox gene A5 (HOXA5)mRNA and its protein in breast cancer tissue,and study the relationship between clinical data and expression of HOXA5 in young breast cancer.Methods:Real-time reverse transcription polymerase chain reaction(Real-time RT-PCR)was used to detect HOXA5mRNA expression in 120 cases of young breast cancer (96 cases of invasive ductal cancer were included) and 96 cases of benign breast lesion.Statistical methods were used to analysis the relationship between clinical data and expression of HOXA5 in young breast cancer.Results:The average expression of HOXA5mRNA in breast cancer tissue was 20.69t29.15 (0.82~189.06),and in benign breast lesions was 31.05±17.06 (5.84~80.72).The expression of HOXA5mRNA was lower in breast cancer than that in benign breast lesions (P<0.01).The expression of HOXA5mRNA was lower in breast cancer with metastatic lymph nodes than that with no metastatic lymph node (P<0.01).The expression of HOXA5mRNA had no relationship with tumor size,clinical stage or histological type of breast cancer (all P>0.05).Conclusion:The expression of HOXA5mRNA in breast cancer tissue is significantly lower than that in benign breast lesions.The low expression of HOXA5mRNA is related with metastatic lymph nodes in young breast cancer.
