超声透析-GFAAS法测定动物组织可溶性小分子蛋白结合铜 被引量:1

Determination of Small Molecular Soluble Protein Binding Copper in Tissue by Graphite Furnace Atomic Absorption Spectrometry after Ultrasonic-Dialysis

摘要 肝豆状核变性、Menke病、婴儿期和儿童期铜中毒综合征、阿尔茨海默病等均与铜代谢失衡有关,如肝豆状核变性疾病患者血清游离铜水平升高、AD病患者脑组织Cu2+与β-淀粉样蛋白(Amyloidβ,Aβ)的异常结合。铜缺乏会导致Cu/Zn—SOD和铜蓝蛋白两种抗氧化成份酶活性降低。 A dialysis-GFAAS method was established for the determination of small molecular soluble protein binding copper (DiaCu) in tissue. 25 kDa dialysis membrane was used, and the dialysate was collected for the copper determination and protein contents determination. The results showed that the detection limit was 0.12 μg/L, with RSD〈5%. The recovery was 90% - 102%. The method was applied to determine the DiaCu in endotoximia SD rats induced by lipopolysaccharide ( LPS), and it was found that the DiaCu concentration decreased in inflammatory tissue.
