摘要 目的观察骨形成蛋白7(BMP7)对环孢素A(CsA)诱导的NRK-52E细胞转分化的影响。方法 NRK-52E经培养传代后随机分为5组:无血清阴性对照组、CsA阳性对照组及BMP7低、中、高剂量干预组(rhBMP7终浓度分别为10,50,100μg/L),每组设5个复孔。24 h后采用Western blot分析各组细胞α-SMA和E-cadherin的表达情况,ELISA法检测各组细胞培养上清液中ColⅠ、ColⅥ浓度。结果 Western blot结果表明,BMP7能剂量依赖性降低α-SMA表达,同时上调E-cadherin表达。结论 BMP7可以逆转CsA诱导的肾小管上皮细胞-间充质细胞转分化,从而减少细胞外基质的生成,发挥肾脏保护作用。 Objective It is to observe the effect of bone morphogenetic protein 7 ( BMP 7) on transdifferentiation of CsA - induced NRK -52E cells. Methods The NRK -52E ceils were divided into five groups after subculture: non-serum negative control group, CsA positive group, and invention groups of low, medium and high doses of BMP 7 (the final concentrations of rhBMP were 10, 50, 100 ug/L respectively) , each group had 5 multiple pores. After 24 h, the expression of ot -SMA and E - cadherin were measured by Western blot, and the levels of Col I and Col V[ were determined by ELISA. Results Western blot results showed that the expression of α - SMA in CsA - treated NRK - 52E cells was up-reglulated, and expression of E - cad- herin was down-regulated. These effects were all reversed by BMP7 dose-dependently. Conclusion BMP7 has a protective effect on NRK - 52E cells induced by CsA, of which the mechanism maybe inhibit the process of EMT.
