摘要
目的考察槲皮素对2型糖尿病大鼠学习记忆能力的影响。方法采用高糖、高脂、高热量饲料预先饲养大鼠并以低剂量链脲佐菌素诱发2型糖尿病大鼠模型,将大鼠随机分为正常组、模型组、槲皮素高、中、低剂量组。采用Morris水迷宫实验考察大鼠的学习记忆能力,并采用电子显微镜观察大鼠海马CA1区突触超微结构的改变,采用免疫组织化学法检测大鼠海马CA1区胰岛素样生长因子Ⅰ(IGF-Ⅰ)含量的变化。结果模型组大鼠在水迷宫的潜伏期较正常组显著延长,在目标象限的停留时间和穿越象限的次数显著减少,海马CA1区内突触结构被破坏,IGF-Ⅰ的含量下降;与模型组相比,槲皮素组大鼠的水迷宫潜伏期显著减短,在目标象限的停留时间和穿越象限的次数显著增加,大鼠海马CA1区内突触结构破坏程度被改善,含量增加。结论槲皮素可改善2型糖尿病大鼠的学习记忆能力,修复突触结构,并增加IGF-Ⅰ的含量。
OBJECTIVE To study the effects of quercetin on diabetes - related rate cognitive impairment. METHODS Diabetes model rats were induced by low - dose streptozotoein after feeded with high - glucose, high - fat and high - calorie feed, and then treated with high, middle and low dosage quercetin. The abilities of learning and memory were tested with Morris water maze. The synaptic ultra- structure in the CA1 region of the hippocampus was observed using electronic microscopy and insulin - like growth factor I in CA1 region of the hippocampus was also determined by immunohistochemistry. RESULTS The diabetes model rats' abilities of learning and memory have been weakened. The contents of IGF - Iin hippocampus CA1 region were deduced and the synaptie structure was destroyed. Compared with model group, quercetin groups treated significantly improved the learning and memory abilities,and ameliorated the content of IGF - I and the structure of synaptic in the CA1 region of the hippocampus. CONCLUSION Quercetin can improve diabetes rats' abilities of learning and memory, and ameliorate the content of IGF - I and the synaptic structure in the hippocampus.
