摘要
目的研究鸢尾苷元在小鼠体内的组织分布,并探讨与中药归经理论的关系。方法用HPLC法测定静脉注射给药后不同时间内小鼠各组织中鸢尾苷元的浓度,流动相为水(磷酸调pH3)-甲醇(42∶58),流速1.0 mL·min-1,柱温35℃,检测波长268 nm。结果鸢尾苷元的各组织匀浆样品在一定浓度范围内线性关系良好,精密度和准确度均符合生物样品测定的要求;小鼠静注2 mg·kg-1鸢尾苷元后,药物在小鼠的心、肝、脾、肾、睾丸、小肠、骨骼肌组织内均有分布,其中以小肠与肝脏组织中分布较多,而肺、脑组织中未检测到药物。结论鸢尾苷元在各组织器官的分布有显著差异,对评价射干及鸢尾苷元的中医归经特点具有重要意义。
OBJECTIVE To study the tissue distribution regularity of teetorigenin in mice and the meridian relations of chinese medi- cine. METHODS HPLC method was used to determine the concentration of tectorigenin in different tissues of mice in different time after iv administration. The mobile phase consisted of water (adjusted to pH = 3.0 by phosphoric acid) -methanol (42 : 58 ) and the flow rate was 1.0 mL. rain-1. The detection wavelength was set at 268 nm and column temperature was 35 ~C. RESULTS Good lineri- ties were obtained in every kind of tissue homogenate sample over a certain range. The precision and accuracy of tectorigenin in tissues met the requirements of biological sample determination. Teetorigenin could be detected in every tissue of the mice, except the lung and brain after intravenous injection of tectorigenin at a dose of 2 mg. kg-1. And the level was higher in intestine and liver than other tissues. CONCLUSION Concentration of tectorigenin has significant difference in different tissues which plays an important role in evaluation of Chinese meridian relational profile of belamcanda chinensis and teetorigenin.
