摘要
目的采用高效毛细管电泳法(HPCE)测定对乙酰氨基酚的含量。方法采用未涂敷熔融石英毛细管柱(50cm×50μm,有效长度45cm)作为分离通道,以50mmol·L-1硼砂(pH9)为缓冲溶液,检测波长243nm,分离电压15kV,虹吸进样,高度差10cm,进样10s,柱温25℃。结果对乙酰氨基酚10~200μg·mL-1与峰面积的线性关系良好(r=0.9991);检测限为1.0g·mL-1(S/N=3),定量限为3.0g·mL-1(S/N=10),加样回收率为97.1%~101.8%(n=3),RSD为0.40%~1.1%。结论所用方法可用于样品中对乙酰氨基酚的含量测定。
OBJECTIVE To establish the method for determination of paracetamol by high performance capillary electrophoresis (HPCE). METHODS The determination was performed with the following conditions : an uncoated fused - silica capillary ( 50 cm × 50 μm ,effective length of 45 cm) ,50 mmol. L-1 borax buffer( pH9 ), a pressure at the injection end with a 10 cm height between the two ends for 10 s. The applied voltage was 15 kV, column temperature was 25 ℃, and detection wavelength was 243 nm. RESULTS The method was validated in terms of accuracy, precision, linearity, stability in solution and proved to be robust. The calibration curves of acetaminophen showed good linearity in the range of 10 - 200 μg mL - 1 ( r = 0.9991 ). The limits of detection ( S/N = 3 ) and quanti- fication ( S/N = 10) were 1.0 μg mL- l and 3.0 μg mL -1 , respectively. The average recoveries at three spiked levels were 97.1% - 101.8% with the RSD of 0.40% - 1.1%. CONCLUSION The method can use for the determination of acetaminophen in the real samples.
