摘要 本文研究了基于高剪切分散乳化技术(High-Speed Shear Dispersing Emulsification,HSDE)提取油橄榄叶中有效成分的样品前处理新方法。利用HPLC测定橄榄苦苷含量,利用UV(AlCl3显色法)测定总黄酮的含量。通过单因素试验分别考察溶剂、转速、料液比、时间及温度,筛选出最佳条件。以浸膏得率、橄榄苦苷和总黄酮的含量为综合指标进行评价,通过响应面中的中心组合设计,优选出最佳提取参数:温度40℃,料液比1∶51.72,溶剂60%乙醇,转速16000 rpm,提取时间150 s。结果表明高剪切分散乳化技术可以快速高效的提取油橄榄叶中有效成分,是一种较好的样品前处理方法。 In this study, a novel pretreatment method based on High-Speed Shear Dispersing Emulsification (HSDE) was developed and optimized for the extraction of active ingredients of Olea europare L.. The contents of oleumpein and total flavonoids were determined by HPLC and UV spectrometer with A1C13 as ehromogenie agent, respectively. The optimum extraction conditions were obtained according to single factor tests. Extraction yield, contents of oleuropein and total fla- vonoids were used as the aggregative indicator for extraction condition optimization in the response surface methodology with central composite design. The optimized extraction conditions were determined as follows: temperature of 40℃, sample to solvent ratio of 1:51.72,60% ethanol as extraction solvent, speed of 16000 rpm, extraction time of 150 s. The results indicated that the HSDE extraction method was efficient for the extraction of active ingredients from olive leaves.
