摘要
为分析翘鳞肉齿菌多糖的抗氧化活性,采用乙醇分级沉淀法分离纯化翘鳞肉齿菌多糖,获得了乙醇体积分数为30%、40%、50%、60%和70%条件下沉析出的5个多糖组分SAP-1、SAP-2、SAP-3、SAP-4和SAP-5;以清除DPPH自由基能力、还原力、清除羟基自由基能力、铁离子螯合能力为指标,评价了5个多糖组分的抗氧化活性。结果表明翘鳞肉齿菌多糖5个组分清除自由基能力、还原力和螯合铁离子能力均表现出一定的浓度依赖性;SAP-4对DPPH自由基的清除活性最高,其EC50值为0.029 g/L,与Vc基本相当;SAP-4对铁离子的螯合能力最强,EC50值为1.208 g/L。
Five polysaccharides fractions ,named SAP-1, SAP-2, SAP-3, SAP-4 and SAP-5, were obtained from Sarcodon aspratus by fractional precipitation with different concentrations of ethanol (volume fraction 30% ,40% ,50% ,60% and 70% ,respectively). The antioxidant activities of polysaccharides were investigated using four different in vitro antioxidant assays, namely 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity assay, hydroxyl radical scaven- ging activity assay, reducing power assay and chelating ability assay. The results showed that DPPH radical scavenging activities, hydroxyl radical scavenging activities, reducing power and chelating ability of the five polysaccharides fractions were dependent on their concentrations. SAP-4 showed the most potent capacity for scavenging DPPH with EC50 values reaching 0.029 g/L,which was similar to the positive control Vc. SAP-4 exhibited the highest capacity for ferrous ions chelating activities with ECso values reaching 1. 208 g/L.
