摘要
目的研究SBA-15对人脐静脉血管内皮细胞(human umbilical vein endothelial cells,HUVEC)的细胞活性和炎症因子C-反应蛋白(CRP)、单核细胞趋化蛋白-1(MCP-1)mRNA表达水平的影响。方法分别将HUVEC细胞暴露于含有终浓度为0(溶剂对照)、25、50、100、200μg/ml的SBA-15和100μg/ml微米二氧化硅(阳性对照)RPMI1640培养基中培养24 h。测定HUVEC增殖的抑制情况、培养液上清中乳酸脱氢酶(LDH)的活力及CRP和MCP-1 mRNA的表达水平。结果与溶剂对照组相比,SBA-15染毒HUVEC的抑制率、培养液上清中LDH活力和CRP和MCP-1 mRNA的表达水平均显著升高;且随着SBA-15染毒浓度的升高,HUVEC的抑制率、培养液上清中LDH活力和CRP和MCP-1 mRNA的表达水平均呈上升趋势。结论 SBA-15对HUVEC具有毒性作用。
Objective To study the effects of SBA-15 on the activity of human umbilical vein endothelial cells (HUVEC) and expression levels of inflammatory factors C-reactive protein(CRP) and monocyte chemotactic factor I(MCP-1) mRNA. Methods HUVEC were exposed to the RPMII640 medium containing a final concentration of 0 (solvent control), 25, 50, I00 and 200 μg/ml of SBA-15 and 100 μg/ml micron silica to be cultured for 24 h. The inhibition of cell proliferation, lactate dehydrogenase (LDH) activity in the medium supernatant and expression levels of CRP and MCP-I mRNA were detected. Results Compared with the solvent control group, the inhibition rate,LDH activity in the medium supernatant and expression levels of CRP and MCP-1 mRNA of ceils exposed to SBA-15 were significantly increased; and with the increase of the SBA- 15 exposure concentration, HUVEC inhibition rate, LDH activity in the medium supernatant and expression levels of CRP and MCP-1 mRNA showed an upward trend. Conclusion SBA-15 has toxic effects on HUVEC.
