摘要
目的 探讨细胞因子信号转导抑制因子2 (SOCS2)和信号转导与转录激活因子5(STAT5) mRNA及其蛋白在肝细胞肝癌中的表达及与临床病理特征的关系.方法 采用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学法检测84例患者肝细胞肝癌、癌旁组织中SOCS2和STAT5 mRNA和蛋白表达水平,并与临床病理资料进行相关分析.结果 肝癌组织及癌旁组织中SOCS2 mRNA相对量分别为0.089±0.034、0.340±0.083,SOCS2 mRNA在肝癌组织中的表达明显低于癌旁组织(P<0.05).肝癌组织及癌旁组织中STAT5 mRNA相对量分别为0.532±0.094、0.276±0.065,STAT5 mRNA在肝癌组织中的表达明显高于癌旁组织(P<0.05).本实验对肝癌组织和癌旁组织中SOCS2和STAT5蛋白表达进行定性分析,在肝癌和癌旁组织中均有两种蛋白的表达,但在肝癌组织中SOCS2的表达明显低于癌旁组织,而STATS在肝癌组织中的表达明显高于癌旁组织.SOCS2和STAT5表达呈负相关,mRNA比较r=-0.612,P<0.05;蛋白比较r=-0.574,P<0.05.结论 在肝癌组织中SOCS2、STAT5 mRNA和蛋白的表达一致,SOCS2在肝癌组织中为低表达,STAT5在肝癌组织中为高表达,两者可能单独或共同参与HCC的发生发展过程.
Objective To investigate the expression of suppressor of cytokine signaling 2 (SOCS2) and signal transducers and activators of transcription 5 (STAT5) in hepatocellular carcinoma (HCC) and their correlation to clinicopathologial features.Methods The mRNA and protein expression levels of SOCS2 and STAT5 in 84 cases of HCC and corresponding paracarninoma tissues were detected by using reverse transcriptase-polymerase chain reaction (RT-PCR) and immunohisochemistry (IHC),respectively.The semiquantitative analysis results and clinical data were statistically analyzed.Results The expression of SOCS2 mRNA in hepatoma tissue and adjacent hepatic tissue to carcinoma respectively was 0.089 ± 0.034 and 0.340 ± 0.083 (P < 0.05).The expression of STAT5 mRNA in hepatoma tissue and adjacent hepatic tissue to carcinoma respectively was 0.532 ± 0.094 and 0.276 ± 0.065 (P < 0.05).The expression of SOCS2 was lower in hepatoma tissue,and that of STAT5 was higher in hepatoma.There was a negative correlation between them in HCC.(r =-0.612,P < 0.01 for mRNA ; r =-0.574,P < 0.01 for protein,respectively).Conclusion There is low expression of SOCS2,and overexpression of STAT5 in HCC.SOCS2 and STAT5 both or alone may participate in the occurrence and development of HCC.
