Expression of c-fos and cjun in cerebral cortex following moderate lateral hydraulic percussion brain injury in rats

摘要 目的 :研究大鼠中度 (0 .2 MPa)侧位液压冲击脑损伤时大脑皮质立即早期基因 c fos和 c jun表达产物 Fos蛋白和 Jun蛋白的变化规律。方法 :雄性 SD大鼠 ,随机分为正常对照组、手术对照组和损伤组。损伤组动物均给以 0 .2 MPa液压冲击脑损伤 ,按冲击后处死时间不同再分为 5、15、30、6 0、12 0、2 40、480和 72 0分钟组。应用免疫组织化学方法观察 Fos和 Jun蛋白在大脑皮质的表达特点。结果 :冲击后 30分钟 ,双侧大脑皮质Fos阳性细胞数逐渐增多 ,冲击后 72 0分钟达高峰。Fos阳性细胞面积在冲击后 12 0分钟逐渐增大 ,72 0分钟达高峰 ;冲击后 6 0分钟 ,双侧大脑皮质 Jun阳性细胞数逐渐增多。冲击后 12 0分钟 ,阳性细胞面积逐渐增大 ,冲击后 72 0分钟 ,阳性细胞数和面积均达高峰。结论 :中度侧位液压冲击脑损伤后 Fos蛋白和 Jun蛋白在双侧大脑皮质均有表达 ,持续时间较长 ,c jun在脑组织内表达的分布范围及变化趋势与 c fos基本一致 ,但表达开始增强的时间较 c Objective:To investigate the protein expression of cfos and cjun in cerebral cortex following moderate lateral fluid percussion brain injury(0 2 MPa) in rats.Methods:Male SpragueDawley rats were divided into normal control,sham operation control and injury group.Animals in injury group were subjected to a moderate lateral hydraulic percussion brain injury (0 2 MPa),and then were subdivided into 5,15,30,60,120,240,480 and 720 minutes groups according to the time elapsed after injury.The expression of cfos and cjun was determined by immunohistochemistry.Results:Thirty minutes after percussion,Fos positive neurons increased bilaterally and progressively in cerebral cortex,peaking at 720 minutes.The area of Fos positive neurons elevated at 120 minutes after percussion,peaking at 720 minutes.Jun positive neurons increased bilaterally at 60 minutes after percussion,and remained elevated up to 720 minutes after brain injury.The area of Jun positive neurons increased at 120 minutes after percussion,peaking at 720 minutes.Conclusions:The induction and expression of cfos and cjun in cerebral cortex increased bilaterally after moderate hydraulic percussion brain injury,and lasted for a long period.The elevated induction of Jun appears to be delayed as compared with Fos,but the temporal pattern and expression extent might be similar to that of Fos.
