摘要
为探讨重组鸡白细胞介素-18 (Interleukin-18,IL-18)与细胞因子诱生剂联合使用对鸡γ-干扰素(Interferon-γ,IFN-γ)和白细胞介素-2(Interleukin-2,IL-2)表达的影响,应用不同浓度的鸡IL-18重组菌和IL-18蛋白与2-脱氧葡萄糖(2-D-G)和植物血凝素(PHA)组成复合制剂,分别经口服、肌肉注射途径免疫雏鸡,检测免疫后不同时间鸡血清中IFN-γ和IL-2含量.结果显示,肌肉注射含重组鸡IL-18蛋白的复合制剂能够明显提高鸡体内IFN-γ和IL-2含量,随IL-18蛋白量的增加,IFN-γ和IL-2含量有升高趋势,且以注射后第7~14天IFN-γ和IL-2含量最高,分别为43 ng/L和70 ng/L,至少在第28天仍能维持较高水平;经口服途径免疫,含鸡IL-18重组菌的复合制剂也能明显提高鸡体内IFN-γ和IL-2含量,免疫后第7天,IFN-γ和IL-2含量最高,分别为50 ng/L和62 ng/L,之后稍有下降,随IL-18重组菌量的增加,IFN-γ和IL-2表达含量变化不大.结果表明,重组鸡IL-18与细胞因子诱生剂组合,经口服或注射途径免疫,均能显著提高鸡IFN-γ和IL-2含量,为新型免疫增强剂和抗病毒制剂的研究和应用提供了参考.
To ascertain the recombined IL-18 with PHA and 2-D-G immunological regulatory of the expression of IFN-γ and IL-2 in chickens,different concentration of IL-18 and PHA as well as 2-D-G were used to immunized unvaccined chicken through oral and intramuscular injection,and ELISA test of IFN-γ and IL-2 expression in chickens.Results showed that intramuscular injection with complex immunopotentiators containing recombined IL-18 protein could induce IFN-γ and IL-2 expression obviously,and ELISA test showed that the expression yield were 43 ng/L and 70 ng/L on 7th day,respectively,and the expression yield decreased slightly after wards.It remained stable till 28th.The different degree of IL-18 in complex immunopotentiators could be observed in regulatory of the expression of IFN-γ and IL-2 in chickens.Meanwhile,immunized through oral with complex immunopotentiators containing recombined IL-18 significantly increased expression of IFN-γ and IL-2,and the expression yield were 50 ng/L and 62 ng/L at 7th,respectively and the expression yield decreased slightly afterwards.Different concentrations of IL-18 in complex immunopotentiators did not increase IFN-γ and IL-2 expression obviously.Results suggested that the complex immunopotentiators could promote IFN-γ and IL-2 contents obviously through both oral and intramuscular injection,which provided reference to the study and application of new kinds of immunopotentiators.
