摘要
取制革厂下水道出口处带有腐烂的动物毛的泥土,在适宜角蛋白酶产生菌的培养基富集培养,得到37株菌。经过固体培养基和发酵培养基复筛,最后从含有腐烂动物毛发的泥土样中筛选得到一株具有较高角蛋白酶活性的菌株,命名为K20。经过革兰氏染色和显微镜观察发现该菌呈杆状,革兰氏染色为阳性。经l6SrDNA序列分析确定该菌株为芽孢杆菌属(Bacillus)。K20菌产生的角蛋白酶用于猪、牛和羊皮脱毛,结果显示该酶具有脱毛活性,且羊皮的脱毛效果较好。
Environmental samples were cultured from the rotten animal hair mud sampled at the outlet of tannery wasterwater, and 37 strains of bacteria were screened and isolated by enrichment culture from soil and sewage. A strain with high keratinase ac- tivity was isolated from the decayed animal hair soil, named K20, and identified by gram - stain reaction, PCR and electron mi- croscope. The results show that the strain is a gram positiye and rod bacteria. The 16SrDNA sequence analysis reveals that the bac- teria is Bacillus. The results of unhairing experiments show that the enzyme come from K20strain has good dehairing properties for cattle hides, pig and sheep skins, especially excellent for sheep skins.
