羌活地黄汤对胶原诱导的大鼠关节炎骨软骨连接的影响 被引量:1

Effects of Qianghuo Dihuang Decoction on the Osteochondral Junction of Collagen-induced Arthritis in Rats

摘要 目的:观察羌活地黄汤对Ⅱ型胶原诱导的大鼠关节炎模型骨软骨连接的影响,探讨羌活地黄汤缓解关节周围骨破坏的作用机制。方法:建立牛Ⅱ型胶原诱导的大鼠关节炎模型,在初次免疫30d后,采用羌活地黄汤灌胃与甲氨喋呤对照,模型组和正常组采用等量的生理盐水灌胃,持续20d后取材,对膝关节骨软骨连接行病理学观察与形态计量分析。结果:关节软骨进行病理学评分,羌活地黄汤组与模型组比较,软骨破坏减轻,差异有统计学意义(P<0.05)。羌活地黄汤组软骨连接处厚度增加,与模型组、甲氨喋呤组比较,差异有统计学意义(P<0.05)。羌活地黄汤组软骨下骨空骨陷窝率减少,与模型组、甲氨喋呤组比较,差异有统计学意义(P<0.05)。结论:羌活地黄汤可缓解胶原诱导的大鼠关节炎模型关节软骨的破坏与减轻关节周围骨侵蚀,其作用机制可能与改善骨软骨连接处的微观结构有关。 Objective.. To observe the effects of Qianghuo Dihuang Decoction on osteochondral junction of collagen-induced arthritis in rats and to investigate the mechanism of alleviating j uxta-articular bone destruction. Methods:Collagen-induced arthritis (CIA) in rats was established. Thirty days after primary immunization, Qianghuo Dihuang Decoction was orally administered for 20 days to rats with CIA. Methotrexate (MTX) was taken as control. Model group and normal group were given the same amount of saline.After sacrifice, pathological observation and morphometry analysis were conducted in osteochondral junction. Results:Compared to model group, there was less damage to the chondrocytes in the Qianghuo Dihuang Decoction treated group. In addition, the subchondral bone plate was almost completely preserved by Qianghuo Dihuang Decoction treatment, while there was a decrease in bone plate integrity in model group. Empty lacunae rate in Qianghuo Dihuang Decoction group showed a lower percentage as compared to the model group and MTX treated rats (P〈0.05). Conclusion:These results reveal that Qianghuo Dihuang Decoction suppresses juxta-articular bone destruction and prevents subchondral bone loss. Its mechanism may be related with improving the microscopic structure of the osteochondral junction.
