摘要
本实验对患病鸭绿江斑鳜进行病毒检测,将表现典型真鲷虹彩病毒(RSIV)病症状的斑鳜鱼肾脏和脾脏研磨成组织匀浆,采用试剂盒方法提取病毒DNA,并应用PCR方法对两对引物进行扩增获得570 bp和568 bp两条基因片段,将其中的568 bp基因片段连接至pMD18-T载体中,经酶切鉴定后序列分析表明,与GenBank登录的RSIV同源性均为99.9%,命名为RSIV-DD。表明本研究在国内首次从淡水鱼斑鳜中分离到RSIV。
Kidneys and livers were collected from the diseased Yalu River mandarin_fish suffered from typical red sea bream iridoviral disease, and grinded into tissue homogenate and the virus DNA was extracted by the kit method and detected by using PCR method. Two gene fragments of 570 bp and 568 bp DNA were amplified by PCR method and the 568 bp DNA was cloned into pMD18-T vector. Sequencing result of the 568 bp DNA indicated that the sequence of nucleotide acid homology was 99.9% with the reference strains of RSIV in GenBank and it was named RSIV-DD. The result demonstrated that the RSIV was isolated for the first time from the naturally infected mandarinfish in China.
