人心房利钠肽对机械通气肺损伤大鼠肺组织NF-κB p65表达的影响

摘要 目的探讨人心房利钠肽(hANP)对大鼠机械通气性肺损伤(VILI)时炎症因子和NF-κB p65表达的影响。方法采用高气道压机械通气模式制备VILI模型。雄性SD大鼠30只随机分为空白对照组(TV组)、机械通气肺损伤组(HV组)和人心房利钠肽组(HV+hANP组)。HV+hANP组于机械通气30 min后静脉注射hANP 0.1 g/(kg·min),机械通气4 h时处死大鼠。免疫蛋白印迹法测定肺组织NF-κB p65的表达;酶联免疫吸附法测定肺组织TNF-α和IL-lβ的含量;计算肺组织湿/干重比(W/D)。结果与TV组相比,HV组、HV+hANP组肺组织TNF-α[(519.22±35.31)、(283.54±24.16)ng/L]和IL-lβ[(751.82±65.35)、(316.29±25.65)ng/L]含量明显上升(P<0.05),NF-κB p65[(238±24)、(178±14)]表达明显增加,肺组织W/D[(9.7±1.2)、(6.4±0.9)]明显升高(P<0.05);与HV组相比,HV+hANP组肺组织TNF-α和IL-lβ含量明显降低(P<0.05),NF-κB p65表达明显抑制,肺组织W/D降低(P<0.05)。结论 hANP通过抑制肺组织NF-κB p65表达,减轻肺部炎症反应,从而减轻大鼠机械通气所致肺损伤。 Objective To investigate the effects of human atrial natriuretic peptide (hANP) on the expression of nuclear factor-kappaB (NF-κB) p65 in the lungs induced by ventilator (VILI) in rats.Methods Rats model of lung injury were induced by ventilation with high peak inspiratory pressure.30 male SD rats were randomly divided into 3 groups,each group had 10 rats:blank control group (group TV),mechanical ventilation lung injury group (group HV) and group HV+ hANP,treated with hANP 0.1 g/(kg· min) after 30 min ventilation.The animals were sacrificed at 4 h after mechanical ventilation.The lungs were immediately removed for determination of expression of NF-κB p65 by Western blot.TNF-α and IL-lβ content were detected by enzymelinked immuno-sorbentassy.Wet to dry weight ratio (W/D) was also calculated.Results Compared with group TV,the expression of NF-κB p65 [(238±24),(178±14)] in the cell nucleus,the level of TNF-α [(519.22±35.31),(283.54±24.16)ng/L],IL-lβ [(751.82±65.35),(316.29±25.65) ng/L] and W/D [(9.7±1.2),(6.4±0.9)] in the lungs significantly increased in group HV and group HV+ hANP (P 〈 0.05); while compared with group HV,they significantly decreased in group HV+ hANP (P 〈 0.05).Conclusion hANP treatment can attenuate the changes in NF-κB p65 expression in the lung induced by ventilation.hANP may alleviate the inflammation of acute lung injury in rat.
