摘要
建立川贝母组培苗快繁体系,为实现川贝母工业化育苗、保护野生植物资源和发展人工育种技术提供新的途径.研究植物激素对川贝母鳞片叶形成愈伤组织、分化小鳞茎和诱导鳞茎抽苗以及无根苗生根的影响,筛选出最佳培养基配方.结果表明,愈伤组织诱导最佳培养基为MS+6-BA 1.0 mg/L+2,4-D 0.5 mg/L,诱导率为84.33%;诱导产生小鳞茎的最佳培养基为MS+KT 2.0 mg/L+NAA 0.3 mg/L,诱导率为82.77%;鳞茎抽苗的最佳培养基为MS+6-BA 3.0 mg/L+IBA 0.3 mg/L,抽苗率为85.39%;无根苗生根最佳培养基为MS+NAA 0.3 mg/L,生根率为83.20%.
The establishment of rapid propagation system of tissue culture of Fritillaria cirrhosa is to provide a new approach of achieving Industrial nursery,protect the wild plant resources and develop the artificial breeding technology.The effect of plant bormone on formation of Fritillaria cirdtosa D.Don Callus,differentiation of bulblets,seedlings induced by pumping the bulb and rooting of rootless seedlings is studied,optimumm medium formula is filtered.The best culture medium of callus induction is MS + 6-BA 1.0 mg/ L + 2,4-D 0.5 mg/L,induction rate is 84.33% ; the best culture medium of bulbs Induction is MS + KT 2.0 mg/L + NAA 0.3 mg/L,induction rate is 82.77% ; the best culture medium of seedlings induced by pumping the bulb is MS + 6-BA 3.0 mg/L + IBA 0.3 mg/L,seeding rate pumping is 85.39% ; the best culture medium of rooting of rootless seedlings is MS + NAA 0.3 mg/L,rooting rate is 83.20%.
