Establishment of focal ischemia-reperfusion model with reforming Longa method in rats

摘要 目的 探索一种创伤小、简便、可重复强的大鼠局灶性脑缺血再灌注模型的制作方法.方法 选取240~280g SD成年大鼠16只,改良线栓法制作大鼠局灶性脑缺血再灌注模型,4、24小时进行神经功能评分,24小时TTC染色检测梗死面积并计算梗死面积/半球面积.结果 4、24小时神经功能评分为1.75±0.68和2.88±1.13;24小时TTC染色示梗死面积/半球面积为(0.45±0.11)%;造模成功率93.75%.结论 该法制作大鼠局灶性脑缺血再灌注模型操作简单、重复性好,效果较理想. Objective To explore and build a reliable rat model of focal cerebral ischemia by a minor trauma and simple operation methods. Method Sixteen SD rats with 240 - 280g were established model by reforming Zea Longa' method. Neurological deficit of rats were graded at 4h and 24h after model building, and the brain sections were stained by TTC ,then calculate the infarct area/brain area. Result Neurological deficit scoring after 4h and 24h was 1.75±0.68 and 2.88±1.13 respectively. TTC show that the infarct area/brain area was(0.45±0.11 ) %. The suc- cessful rate of execution was 93.75%. Conclusion The modified focal ischemic model in rats is an ideal animal model with simple operation and higher reproducibility.
