摘要
建立苯酚-硫酸法测定黄河口沉积物中的多糖,确定了最佳实验条件:最大吸收波长为487nm,苯酚质量分数为12%,95.5%的浓硫酸用量为4mL,溶液反应体积为2.5mL,绘制标准曲线所采用的标准物质为含有0.1g沉积物空白样品和一定量葡萄糖的混合物。结果表明,在优化实验条件下,成功获得多糖与葡萄糖之间的换算因子为4.85。该方法的精密度为4.4%,平均回收率为98.68%,相对标准偏差为2.3%。应用本方法测出黄河口沉积物中的多糖平均含量为0.1%,相对标准偏差为2.8%,与有关文献报道相符合,表明该方法能够满足黄河口沉积物中多糖含量测定的要求。
The phenolsulfuric acid method has been developed for the determination of polysaecharides in sediment at the Yellow River Estuary. The optimum experimental conditions were established, including maximum absorption spectra was 487 nm, phenol mass fraction was 12%, 95.5% sulfuric acid volume was 4 mL, solution reaction volume was 2. 5 mL. The standard curve was drew by selecting the standard material consisting of glucose and 0. lg sediment blank sample. The results showed that under the optimized conditions the conversion factor between polysaccharides and glucose was 4. 85, the precision of this meth od was 4.4 %, the average recovery rate was 98.68 %, RSD was 2.3 %. By this method the measured aver age content of polysaccharides in Yellow River Estuary sediment was 0. 1%, RSD was 2.8%. This was consistent with related literatures, it was indicated that this method could be successfully applied to the detection of polysaccharides in sediment at the Yellow River estuary.
