摘要
目的:探讨百事乐胶囊对慢性应激抑郁大鼠海马神经元形态结构和功能的影响。方法:在48只SD大鼠中随机选择12只(雌雄各半)为正常组,不进行任何刺激,正常饲养。对其余大鼠给予21d慢性不可预见性应激造模,造模完成后将其随机分为模型组、百事乐组和氟西汀组,每组12只,雌雄各半。对各组大鼠予相应药物进行灌胃给药治疗,模型组大鼠灌胃蒸馏水,每天1次,灌胃给药1h后,再随机给予1种刺激,持续21d。分别于造模后及给药结束后测定各组大鼠糖水偏好度,尼氏染色观察大鼠海马神经元尼氏体,免疫组化检测大鼠海马MAP-2的表达。结果:与模型组相比,百事乐组和氯西汀组大鼠的糖水偏好度明显提高(P<0.01);模型组大鼠海马尼氏体减少,颜色变浅,而百事乐组大鼠尼氏体丰富,胞质色深;免疫组化显示百事乐胶囊可提高抑郁大鼠海马MAP-2的平均光密度值,与模型组比较,差异有统计学意义(P<0.01)。结论:百事乐胶囊可通过增加尼氏小体的数量,调节海马MAP-2的表达以促进抑郁大鼠海马神经功能恢复。
Objective : To investigate the effect of Baishile capsules on the morphological structure and function of hippocampal neurons in rats with depression induced by chronic stress. Methods:Twelve rats( normal group, male -to -female ratio = 1:1 )were randomly se- lected from 48 Sprague - Dawley rats to be normally fed without stimulation. Other rats were treated with chronic unpredictable stress for 21 d to establish a rat model of depression, and the treated rats were randomly divided into model group( n = 12), Baishile group( n = 12) ,and fluoxetine group(n = 12) ,with a male - to - female ratio of 1 : 1 in each group;the Baishile group and fluoxetine group re- ceived Baishile and fluoxetine by garage, while the model group was given distilled water by gavage ; a randomly selected pattern of stim- ulation was performed at 1 h after intragastric administration,and this treatment was performed once daily for 21 d. After the model was established and intragastric administration was performed, sucrose preference test was conducted, the Nissl bodies in hippocampal neu- rons were observed by Nissl staining, and the hippocampal microtubule -associated protein -2 (MAP -2)expression was measured by immunohistochemistry. Results:Compared with the model group, the Baishile group and fluoxetine group had significantly increased su- crose preference ( P 〈 0.01 ). In the model group, the number of Nissl bodies in hippocampal neurons was reduced, and their color be- came lighter;in the Baishile group, there were abundant Nissl bodies, and the cytoplasm was dark- colored. The immunohistochemistry showed that the Baishile group had significantly increased mean optical density of hippocampal MAP - 2 compared with the model group (P 〈 0. 01). Conclusion :Baishile capsules can increase the number of Nissl bodies and upregulate hippocampal MAP -2 expression to promote the functional recovery of hippocampal neurons in rats with depression.
