左金丸对应激性胃溃疡大鼠作用机制的研究

Study on action mechanism of Zuojin pills in rats with stress- induced gastric ulcer

目的:探讨左金丸对因"束缚—水浸"应激引起的大鼠应激性胃溃疡胃黏膜损伤的保护作用及机制。方法:将60只健康SD雄性大鼠随机分为空白对照组,模型对照组,阳性对照组,左金丸低、中、高剂量组,每组10只。各组预防性给药,空白对照组和模型对照组大鼠均灌胃生理盐水,阳性对照组灌胃奥美拉唑水悬液,左金丸低、中、高剂量组分别灌胃左金丸低、中、高剂量水煎剂,连续5d。末次给药后禁食不禁水24h,采用"束缚—水浸"应激法制备大鼠应激性胃溃疡模型。造模成功后进行胃体形态学观察,测量胃液PH值,计算胃溃疡指数(UI),采用酶联免疫法检测血清中促肾上腺皮质激素(ACTH)、皮质酮(CORT)的含量。结果:左金丸组大鼠胃液PH值明显升高,UI显著降低,血清ACTH、CORT水平显著下降,与模型对照组比较,差异均具有统计学意义(P<0.05,P<0.01)。结论:左金丸可以预防应激性胃溃疡的发生并能促进胃溃疡的愈合,其作用机制可能与提高胃液PH值从而降低胃黏膜攻击因素有关,同时通过内分泌调节胃黏膜防御因子与攻击因子失衡,也是其作用机制之一。

Objective: To investigate the protective effect of Zuojin pills in rats with stress- induced gastric ulcer caused by restraint-water immersion and the mechanism by which Zuojin pills protect the rats from gastric mucosal injury. Methods: Sixty healthy male Sprague- Dawley rats were randomly divided into blank control group,model control group,positive control group,and low-,middle-,and high- dose Zuojin pill groups. The rats received respective treatments by prophylactic administration for 5 d: the blank control group and model control group received normal saline by gavage; the positive control group received water suspension of omeprazole by gavage; the low-,middle-,and high- dose Zuojin pill groups received low-,middle-,and high- dose water decoction of Zuojin pills by gavage. After the last administration,fasting( without water deprivation) was performed for 24 h,and a rat model of stress- induced gastric ulcer was established by restraint- water immersion. Gastric morphology was observed; the pH value of gastric juice was measured; gastric ulcer index( UI) was calculated; serum levels of adrenocorticotropic hormone( ACTH) and corticosterone( CORT) were measured by enzyme- linked immunosorbent assay. Results: The Zuojin pill groups had significantly increased pH value of gastric juice,significantly decreased UI,and significantly decreased serum levels of ACTH and CORT compared with the model control group( P <0. 05 or P <0. 01). Conclusion: Zuojin pills can prevent the development of stress- induced gastric ulcer and promote the healing of gastric ulcer. The action mechanism of Zuojin pills may include reducing the aggressive factors for gastric mucosa by increasing the pH value of gastric juice and reducing the imbalance between defensive factors and aggressive factors by endocrine regulation.
